LB996 Lipid droplets are a metabolic vulnerability in melanoma
نویسندگان
چکیده
Melanoma cells exhibit numerous transcriptional cell states including developmental programs such as neural crest-like and pigmented melanocytic cells. These endow tumorigenic properties like sustained growth, immune evasion, therapy resistance, metastasis. Here, we use a zebrafish melanoma model to identify program linking the state dependence on lipid droplets, specialized organelle responsible for storage. Single-cell RNA-sequencing of these tumors show concordance between genes regulating pigmentation those involved in oxidative metabolism. This metabolic phenotype is conserved human specimens. demonstrates increased fatty acid uptake, an number upon Genetic pharmacologic suppression droplet production sufficient disrupt metabolism slow growth vivo. Because linked poor outcomes patients, data indicate vulnerability that depends organelle.

Some Lipid Droplets Are More Equal Than Others: Different Metabolic Lipid Droplet Pools in Hepatic Stellate Cells
Hepatic stellate cells (HSCs) are professional lipid-storing cells and are unique in their property to store most of the retinol (vitamin A) as retinyl esters in large-sized lipid droplets. Hepatic stellate cell activation is a critical step in the development of chronic liver disease, as activated HSCs cause fibrosis. During activation, HSCs lose their lipid droplets containing triacylglycerol...
متن کاملLipid Droplets as Signaling Platforms Linking Metabolic and Cellular Functions
The main cells of the adipose tissue of animals, adipocytes, are characterized by the presence of large cytosolic lipid droplets (LDs), which store triglyceride (TG) and cholesterol. However, most cells have LDs and the ability to store lipids. LDs have a well-known central role in storage and provision of fatty acids and cholesterol. However, the complexity of the regulation of lipid metabolis...

OK, they are cute, but why should a developmental biologist care about them? Onset of calcification early in ontogeny coupled with gradual, progressive morphological development permits the recognition of growth sequences for over a hundred trilobite species. Striking variation in the number of trunk segments at maturity across ontogeny and phylogeny, coupled with the curious pattern of segment...

متن کاملMature lipid droplets are accessible to ER luminal proteins.
Lipid droplets are found in most organisms where they serve to store energy in the form of neutral lipids. They are formed at the endoplasmic reticulum (ER) membrane where the neutral-lipid-synthesizing enzymes are located. Recent results indicate that lipid droplets remain functionally connected to the ER membrane in yeast and mammalian cells to allow the exchange of both lipids and integral m...
